Yes, but if you measure from the same edge, you'll get two inches, not one (assuming the figure has a one-inch base).

For example, assume this is your map. The A is where you measure from, the B is the base of the mini, and four characters make up one inch:

____ABBB____ABBB____

There's one inch between the two bases, but since you measure from the A to the A you get two inches between the two minis.
