No. However it does depend on what you are expecting this LOCK to do. If you are expecting it to hold fast to the solid ground, it won't work. Yes. Assuming you are LOCKing the flip top or the strings, etc. that closes or opens a portal.

LOCKing the pouch portal will make it harder to open, not harder to take off. If the pouch was an integral part of a belt, then casting it on the buckle would prevent the buckle from opening.
Can you wrap a rope around a prisoner's hands then cast the Lock spell to bind their hands?
By wrapping a rope around, you mean tying them closed? Yes, I would say it would LOCK the knot.

Would this prevent anybody from lifting your hidden dagger? Especially with five levels of conceal on it?
No. I'm assuming that the dagger is in the boot. As such, it can be pulled out if it can be found.
To me, a LOCK, needs to be put on a common concept portal access item that can prevent access: A door, a gate, a drape.
The farther it gets from this "commonly accepted concept" the more difficult it is for the wizard to believe that it really fits the concept. It should either cost more ST to empower, higher DX (more dice?) to succeed, or greater wisdom (-x to IQ) to believe or maybe combinations to do it.
Then again, it does talk about a door not moving if you intend to hold it closed. So maybe it is the entire object and not just the lock or knot. heh.
