"The Last Laugh" card

I have been playing Munchkin for some time and just recently picked of the Gets Promoted pack. We played with the Last Laugh card and we have a question. In Munchkin Quest you can kill your character at any time just by turning over your heart tokens. Can you kill your character at any time in the Munchkin card games in order to use this card for the winning level? It says you can use it as the winning level, we just aren't sure about if you can kill your character at any time to achieve this goal. Thanks
