Does the +3 against males go to +6 when another male helps kill it?

No. The bonus applies if there is any male in the battle (main combatant, helper, or Hireling/Comrade) and does not stack if there are multiple ones. Such bonuses only apply once per thing referenced (and the only thing referenced by Marique is males).
