I see your confusion.
Does the monster get its +3 bonus if either of the munchkins has no big item?
In other words, one of them has a big item, but the other does not.
The wording would suggest that it does, since the one munchkin satisfies the condition that it has no big item.

If at least one munchkin in the fight lacks a Big item, the Rune Giant gets +3. The bonus does not stack if both lack a Big item.

UPDATE: The Rune Giant reads "+3 against anyone with no Big items." So I'm reversing the previous ruling to this extent: if two munchkins are fighting it and neither one has a Big item, the Rune Giant is at +6. It's not a case where it's just "+3 against Elves"; the wording here is more specific than that.

Similarly, (and you can move this from 101 if need be,) does the Kevinator from the Guild booster (+5 against Everyone) get another bonus against a helper?

No, because "everyone" isn't synonymous with "anyone." Kevinator gets a flat +5 bonus.

I'd argue the grammar on that the other way around myself. Add back the spaces - against any one gives you a bonus against one person, against every one would be a separate bonus against each of however many there were.
