Default (DF) Best class for sneaking up on someone and killing them?

What do you reckon the best DF class is for sneaking up on a foe and killing them at close range?

Ninja? Assassin? Monk? Mage? Other?

Depends on the details of what you're sneaking up on; some classes are better at the 'sneak up' part, others at the 'kill' part.

Assassin. (DF12), (P#3:50 for Power-Ups)

Grab as Advantages: Night Vision, Striking ST (only on surprise attacks) and Luck.

As Power-Ups: Backstabbing (for when stealth fails), Silence, more Striking ST on surprise attacks, Weapon Master, Focused Fury if you more often need to down the target to start a fight, rather than down it silently and move forward, Bane-Brewer perk if you want to Bladeblack the most important targets.
The book recommends Trademark backstabbing moves. I have created one that focus on looking cool, keeping the target silent and dealing quite some damage (it's not very optimized for damage, though), while staying strictly by the books.

However, that uses some Martial Arts rules and your GM might want to restrict you on how much of the penalties you can buy off, sticking to Slayer Training(DF11).

So a Wizard with Invisibility and Mage Stealth is extremely difficult to detect via mundane means, they may not be the best to deliver the kill though. I guess a Wizard that buffs the Barbarian to undetectably axe people in the head is a good combo but maybe not in the spirit of the original question.

3d6 Deathtouch is pretty damn nasty.

But if the target lives (or worse shakes it off little worse for the wear) then the Wizard is boned.

Barbarian. Half-Elf Barbarian if you want to really specialize. Decent (for Elf, very good) stealth, and a telegraphic all out to the skull at a Barbs ST is a death blow on anything WITH a skull. Even without a hit location, 3d-1 swing should drop anything meant to be dropped (non-boss monsters). Elf Barbs make good snipers too.

And if the first blow effs up somehow, the Barbarian is extremely well suited to dealing with the aftermath.

The shadow elf ninja in my game was really effective at this.

I think it's relevant though - parties that work together and help each other do better than parties that act as 5 solo units.

Usually in the party with ninja the bard would distract them (one way or another) while the ninja killed them...

9d missile spell at point-blank range

18d missile at point-blank, with Magery 6

Being an elf wizard with Magery 9, a 27d missile spell...like Stone Missile to the skull.
