The gentleman in the clip looks over some 4-bore rifles at auction and takes one to the range to fire at the end of the video.
Those would be fired using the Cannon skill, by the way. Sweet canola that's a lot of gun.

I like how the narrator winces a little bit when he describes the bullet as a quarter pound of lead.

No. Guns (Rifle). Compare High-Tech: Adventure Guns, p. 35. The largest listed there is 8-bore, but that doesn't change anything. These do not use Gunner (Cannon).

It's a very fine line for AMRs firing 20mm rounds as to whether they're classified in real life as Rifles or Cannon, as 20mm tends to be the cut-off.

In GURPS, I'd say it comes down to whether it's fired like a rifle from the prone position with a bipod, in which case it's Guns (Rifle), or if the thing is mounted like on an aircraft or tank, which would make it Gunner (Cannon). (Autofire versions would be Guns (LMG) or Gunner (MG), respectively.) Mounted on a tripod... that's a gray area, which I'd rule it could use the better of either skill.

Calibre is of very limited usefulness to find which rule is required. Basically, everything mounted is Gunner, everything fired from the shoulder is Guns. Both the LAR Grizzly Big Boar and the old 4-bore rifles use Guns (Rifle).

Oh, yes. The punt gun. It's a 1-gauge shotgun used to turn an entire flock of waterfowl into instant paté.

Tripods are normally Gunner.

Maybe for a GPMG that's stocked and usually used with Guns (LMG), which is occasionally mounted on a small tripod, using Guns with the tripod might be appropriate.

I'm surprised this is even debated. We laid out the rules quite clearly in High-Tech (p. 129).
