I usually roll playmats semi-loose. Nothing too tight, but I have no evidence that one is better than the other. In my experience, when boxed or tubed, they'll unroll slightly to fill the space.
I also roll them with the neoprene side facing out. Again, I have no evidence that one way is better than the other.
