There is no stack, nor 'in response' in Munchkin. Once an action starts, it must be completed before you take the next action, such as playing a card. Thus, once the Thief discards his card in payment for the backstab, the target Munchkin is backstabbed.

Except, of course, the cards which actually are designed to be played in response to something, like
Wishing Ring, which can be played in response to a Curse to prevent the Curse from ever happening. . .
